[2020 TTS] Three research results jointly conducted by clinical experts and AlloDx will be presented at the TTS online annual meeting

Affected by the global new coronavirus epidemic, the event was originally scheduled to be held in Seoul, South KoreaThe 28th World Transplantation Congress (TTS)Change toOnline meeting format, the meeting time is September 13-16, 2020 (US Eastern Time). This conference invited more than 5,000 experts in the field of transplantation from 105 countries/regions. The three-day conference schedule includes plenary sessions, special seminars, TTS branch meetings, oral speeches, etc., totaling250 reports, covering multiple hot topics in the field of transplantation.

Simultaneously detection of ddcfDNA in plasma and urine assist to determine the type of rejection in renal transplant patients [Oral Presentation]

The Kidney Disease Center of the Eastern Theater Command General Hospital and AlloDx collaborated to study the role of ddcfDNA (donor-derived cell-free DNA) in the plasma and urine of kidney transplant patients in identifying and diagnosing rejection injury types. This research will be shared with participants in the form of Oral Presentation [7:30 am on September 15, Beijing time].

This study enrolled 56 kidney transplant patients with acute rejection confirmed by biopsy, and used the Target Region Capture Sequencing method to detect the relative and absolute values of ddcfDNA in the patients' plasma and urine. The results showed that there was a significant difference in plasma ddcfDNA between the TCMR IA/IB group and the ABMR group (p=0.013) and the TCMR IIA/IIB group (p=0.01). Urinary ddcfDNA has significant differences among the three groups of ABMR, TCMR IA/IB and TCMR IIA/IIB (p<0.001; p<0.001; p=0.047), and the fragment length distribution of ddcfDNA has auxiliary diagnostic significance for ABMR.

The level of plasm donor-derived cell-free DNA in kidney transplant patients with severe pneumonia [Poster]

The results of the collaborative research between transplant experts from the Second Xiangya Hospital of Central South University and AlloDx will be displayed in the form of posters. This study enrolled 55 patients, divided into infection group (13 cases, before treatment), remission group (13 cases, on the 7th day of treatment) and stable group (42 cases). The results suggest that severe pneumonia can cause transplanted kidney damage and increase the absolute value of ddcfDNA in serum, but the relative value does not change significantly. The relative value of serum ddcfDNA may be diluted by the release of cfDNA (recipient cfDNA) from lymphocyte apoptosis and necrosis. Therefore, the relative value of ddcfDNA is used to monitor rejection, which is not affected by pneumonia.

Noninvasive genotyping of donor’s HLA in kidney transplant patients by target capture sequencing [Poster]

AlloDx is working with transplant experts to study donor HLA typing via urine ddcfDNA, which will be presented as a Poster. The results show that when the ddcfDNA concentration exceeds 25%, donor HLA can be stably typed. This study provides a new detection method for recipients whose donor HLA type is unknown.
