- Research ideas:This study performed whole-genome methylation sequencing analysis on 12 transplanted kidney tissues before ischemia (before transplantation), after reperfusion, 3 months after transplantation and after transplantation, in order to find the changes in gene methylation levels of transplanted kidney tissue caused by ischemia and reperfusion.

Research results:
(1) All transplanted kidneys caused an increase in tissue methylation after ischemia, and the longer the ischemia time, the more obvious the increase in methylation. For every 1 hour of ischemia time, the methylation level increased by 2.6%.
(2) The degree of methylation on the genome increases in more biased regions, mostly in genes related to inhibiting kidney damage and fibrosis, that is, the methylation levels of these genes are inhibited.
(3) Hypermethylation of genomic CpG islands in transplanted kidneys before transplantation predicts chronic injury, abnormal fibrosis and glomerulosclerosis 1 year later.
